Hydrophilic Pd0 complexes based on sugars for efficient Suzuki-Miyaura coupling in aqueous systems

Two classes of hydrophilic Pd0 complexes containing P,N and N,N sugar-based ligands were prepared and tested in the Suzuki-Miyaura cross-coupling reaction under environmentally friendly aqueous conditions. The best catalyst was tolerant towards different substrates, and its activity is comparable with the highest values reported so far for reactions in aqueous media [loading 0.0010 %, turnover frequency (TOF) 3.5 ¡Á 104 h-1].

Evidence for a Cooperative Mechanism Involving Two Palladium(0) Centers in the Oxidative Addition of Iodoarenes

Oxidative addition of iodoarenes (ArI) to Pd0 ligated by 1-methyl-1H-imidazole (mim) in polar solvents leads to cationic arylpalladium(II) complexes [ArPd(mim)3]+. Kinetic analyses evidence that this reaction is second order with respect to the concentration of Pd0, and a mechanism involving the cooperative intervention of two Pd0 centers has been postulated to explain this finding. This unusual behavior is also observed with other nitrogen-containing ligands and it is general for iodobenzenes substituted with electron-donating or weakly electron-withdrawing groups. In contrast, bromoarenes and electron-poor iodoarenes display first-order kinetics with respect to Pd0. Theoretical calculations performed at the density functional theory (DFT) level suggest the existence of mim-ligated ArI-Pd0 complexes, in which the iodoarene is bound to the metal in a halogen-bond-like fashion. Coordination weakens the C?I bond and facilitates the oxidative insertion of another Pd0 center across this C?I bond. This conceptually novel mechanism, involving the cooperative participation of two distinct metal centers, allows a full explanation of the experimentally observed kinetics.

Suzuki-Miyaura Diversification of Amino Acids and Dipeptides in Aqueous Media

The Suzuki-Miyaura derivatisation of free amino acids, peptides and proteins is an attractive area with considerable potential utility for medicinal chemistry and chemical biology. Here we report the modification of unprotected and Boc-protected aromatic amino acids and dipeptides in aqueous media, enabling heteroarylation and vinylation. We systematically investigate the impact of the peptide backbone and adjacent amino acid residues upon the reaction. Our studies reveal that although asparagine and histidine hinder the reaction, by utilising dppf, a ferrocene-based bidentate phosphine ligand, cross coupling of halophenylalanine or halotryptophan adjacent to such a residue could be enabled. Our studies reveal dppf to have good compatibility with all unprotected, proteinogenic amino acid side chains.

Palladium-Catalyzed Arylation of Arylboronic Acids with Yagupolskii-Umemoto Reagents

A Pd-catalyzed Suzuki cross-coupling of arylboronic acids with Yagupolskii-Umemoto reagents was explored. In contrary to trifluoromethylations, the Pd-catalyzed reaction of R-B(OH)2 and [Ar2SCF3]+[OTf]- provided the arylation products (R-Ar) in good to high yields. The reaction confirms that the S-Ar bonds of [Ar2SCF3]+[OTf]- can be readily cleaved in the presence of Pd complexes. The relatively electron-poor aryl groups of asymmetric [Ar1Ar2SCF3]+[OTf]- salts are more favorably transferred compared to the electron-rich ones. This reaction represents the first report of utilization of [Ar2SCF3]+[OTf]- as arylation reagents in organic synthesis.

The product ratio is dependent on the choice of the solvent and electronic nature of the aromatic substituent.

We report a new protocol for the annulative difunctionalization of acetylenes via tandem carbocyclization-coupling of ?-acetylenic beta-ketoesters with aryl and heteroaryl bromides and chlorides catalyzed by the palladium species derived from an air- and moisture-stable palladacyclic precatalyst. In the tandem process, the palladium complex combines appropriate carbophilic Lewis acidity and redox activity to catalyze two mechanistically distinct reactions – nucleophilic addition of the enolate to unactivated alkyne, followed by C-C coupling. We found that a broad range of electronically varied aryl and heteroaryl bromides and chlorides underwent this reaction with various ?-acetylenic beta-ketoesters, providing corresponding substituted vinylidenecyclopentanes in high yield with excellent functional group tolerance.
