Facile synthesis of beta-derivatized porphyrins – Structural characterization of a beta – beta-bis-porphyrin

A generally applicable Suzuki methodology for the synthesis of beta- derivatized porphyrins has been developed starting from bromoporphyrin 1, which is converted into an air- and water-stable boronate derivative. Metal- mediated cross-coupling of the latter compound with various aryl bromides furnishes beta-arylporphyrins 2 with yields similar to those reported for derivatization at the meso-position. The utility of the method was demonstrated with the preparation and structural characterization of bis- porphyrin 3, directly linked at the beta-positions of the rings.

They are accessible by diverse consecutive reaction sequences, which allow the introduction of transition metal units such as Fc, [(tBu2bpy)(CO)3Re], [(eta5-C5H5)(Ph3P)2Ru], [(eta5-C5H5)(Ph3P) 2Os], and trans-[(Ph3P)2(Cl)Pt] (Fc = (eta5-C5H5)(eta5-C 5H4)Fe; tBu2bpy = 4,4?-di-tert-butyl-2,2?-bipyridyl). The solid state structures of five complexes have been determined. The electrochemical behavior of the newly synthesized mono-, heterobi-, and heterotrimetallic assemblies have been studied, showing that there is no significant electronic interaction between the respective metal atoms. UV-vis spectroscopic measurements suggest a weak interaction between the appropriate metal atoms. The associated radical cations were in situ generated by stepwise chemical oxidation and characterized by continuous wave electron paramagnetic resonance (EPR) investigations in X-band performed at low temperatures.

A palladium-catalyzed domino Heck/silylation reaction of acrylamides with hexamethyldisilane is presented. This transformation, which involves the formation of a sigma-alkylpalladium intermediate, allows direct access to functionalized (2-oxoindolin-3-yl)methylsilanes in good to excellent yields. Furthermore, this strategy could be utilized for the synthesis of (2-oxoindolin-3-yl)methylstannanes from hexamethyldistannane. (Figure presented.).

Benzoyl Fluorides as Fluorination Reagents: Reconstruction of Acyl Fluorides via Reversible Acyl C-F Bond Cleavage/Formation in Palladium Catalysis

This report describes the formation of value-added acyl fluorides by means of palladium-catalyzed acyl-exchange reactions between acyl fluorides and acid anhydrides. This method allows using a simple and commercially available acyl fluoride, benzoyl fluoride, as the fluoride source for the easy and efficient preparation of a variety of more complex acyl fluorides. The results of this study suggest that this reaction proceeds via a reversible acyl C-F bond cleavage/formation at the palladium center.

Discovery of 6-[5-(4-fluorophenyl)-3-methyl-pyrazol-4-yl]-benzoxazin-3-one derivatives as novel selective nonsteroidal mineralocorticoid receptor antagonists

In the course of our study on selective nonsteroidal mineralocorticoid receptor (MR) antagonists, a series of novel benzoxazine derivatives possessing an azole ring as the core scaffold was designed for the purpose of attenuating the partial agonistic activity of the previously reported dihydropyrrol-2-one derivatives. Screening of alternative azole rings identified 1,3-dimethyl pyrazole 6a as a lead compound with reduced partial agonistic activity. Subsequent replacement of the 1-methyl group of the pyrazole ring with larger lipophilic side chains or polar side chains targeting Arg817 and Gln776 increased MR binding activity while maintaining the agonistic response at the lower level. Among these compounds, 6-[1-(2,2-difluoro-3-hydroxypropyl)-5-(4-fluorophenyl)-3-methyl-1H-pyrazol-4-yl]-2H-1,4-benzoxazin-3(4H)-one (37a) showed highly potent in vitro activity, high selectivity versus other steroid hormone receptors, and good pharmacokinetic profiles. Oral administration of 37a in deoxycorticosterone acetate-salt hypertensive rats showed a significant blood pressure-lowering effect with no signs of antiandrogenic effects.

Quantifying the Interdependence of Metal?Ligand Covalency and Bond Distance Using Ligand K-edge XAS

Bond distance is a common structural metric used to assess changes in metal?ligand bonds, but it is not clear how sensitive changes in bond distances are with respect to changes in metal?ligand covalency. Here we report ligand K-edge XAS studies on Ni and Pd complexes containing different phosphorus(III) ligands. Despite the large number of electronic and structural permutations, P K-edge pre-edge peak intensities reveal a remarkable correlation that spectroscopically quantifies the linear interdependence of covalent M?P sigma bonding and bond distance. Cl K-edge studies conducted on many of the same Ni and Pd compounds revealed a poor correlation between M?Cl bond distance and covalency, but a strong correlation was established by analyzing Cl K-edge data for Ti complexes with a wider range of Ti?Cl bond distances. Together these results establish a quantitative framework to begin making more accurate assessments of metal?ligand covalency using bond distances from readily-available crystallographic data.

An efficient approach for direct carbamoylation of terminal alkynes with formamides affording propiolamides has been developed by copper-catalyzed oxidative cross coupling of C(sp)-H and C(sp2)-H bonds in the presence of a pincer ligand with two imidazolyl groups. The catalytic reaction is compatible with diverse functional groups but sensitive to the electronic effect of terminal alkyne and the steric effect of formamides. KIE study indicates the cleavage of the carbamoyl C-H bond affording formamide radical is the rate-determining step.
