A diastereoselective Pd-catalyzed intramolecular cyclopropanation of alkenes with unstabilized allylic tosylhydrazones was developed. This methodology provides an efficient entry toward synthesis of the bicyclo[3.1.0] hexane system with an exo-double bond, and sets the basis for future elaboration of more complex polycyclic motifs.

Discovery of biphenyl-aryl ureas as novel VEGFR-2 inhibitors. Part 4: Exploration of diverse hinge-binding fragments

Abstract VEGFR-2 plays an essential role in angiogenesis and is an important target for cancer therapy. A series of biphenyl-aryl ureas were synthesized and evaluated as novel VEGFR-2 inhibitors. The pyridine, methylamine carbonyl pyridine and pivaloyl amide pyridine were introduced as novel hinge binding fragment. The majority of title compounds displayed potent VEGFR-2 inhibition. In particular, L1, L9, W14 and W15 exhibited significant enzymatic inhibitory activity with IC50 values of 0.36 nM, 0.22 nM, 0.15 nM and 0.14 nM. Compounds L1, L9 and W15 displayed potent antiproliferative activity against A549 and SMMC-7721 cells. SAR study suggested that incorporation of 3-trifluoromethyl and methylamine carbonyl on terminal pyridine could improve VEGFR-2 inhibitory activity. Molecular docking illustrated that urea moiety formed two critical hydrogen bonds with the DFG residues of VEGFR-2. The results indicated that these biphenyl-aryl ureas could serve as promising lead compounds for further optimization.

Synthesis and near infrared electrochromic properties of metallodithiolene complexes

Four metallodithiolene complexes [4,8-bis(octyloxy)-1,3,5,7-tetrathia]?di[1,1?-bis(diphenylphosphino)ferrocene?palladium(II)] (3), [4,8-bis(octyloxy)-1,3,5,7-tetrathia]di[1,3-bis(diphenylphosphino)propane?nickel(II)] (4), [4,8-bis(octyloxy)-1,3,5,7-tetrathia] ?[1,1?-bis(diphenylphosphino)ferrocene?palladium(II)]?[1,3-bis(diphenylphosphino)propane·nickel(II)] (5) and di[4,8-bis (octyloxy)-1,3,5,7-tetrathia]?[1,1?-bis(diphenylphosphino)ferrocene?palladium(II)]?nickel(II) (6) were synthesized and the near-infrared (NIR) electrochromic properties were studied. The spectroelectrochemical spectra and the electrochromic parameters such as optical contrast, switching time, optical density change, electrochromic efficiency and optical attenuation of complexes 3?6 were investigated in detail. The symmetric binuclear complex 4 showed relatively high electrochromic efficiency of 63.0 and 75.4 cm2/C both in the two oxidation states. The complexes exhibited excellent electroactive/electrochromic stability characterized by chronoamperometry (>4000 cyclic switches).

In particular, the nature of the surfactant has been found to be crucial in order to obtain synthetically useful results involving highly reactive, moisture-sensitive organometallics. Effect of Perylene Photosensitizer Attachment to [Pd(triphosphine)L]2+ on CO2 Electrocatalysis

Two new covalently linked chromophore-CO2 reduction catalyst systems were prepared using a perylene chromophore and a bis[(dicyclohexylphosphino)ethyl]phenylphosphinopalladium(II) catalyst. The primary goal of this study is to probe the influence of photosensitizer attachment on the electrocatalytic performance. The position either para or meta to the phosphorus on the phenyl group of the palladium complex was linked via a 2,5-xylyl group to the 3 position of perylene. The electrocatalytic CO2 reduction activity of the palladium complex is maintained in the meta-linked system, but is lost in the para-linked system, possibly because of unfavorable interactions of the perylene chromophore with the glassy carbon electrode used. Following selective photoexcitation of the perylene, an enhanced perylene excited-state decay rate was observed in the palladium complexes compared to perylene attached to the free ligands. This decrease is accompanied by formation of the perylene cation radical, showing that electron transfer from perylene to the palladium catalyst occurs. Electron transfer and charge recombination were both found to be faster in the para-linked system than in the meta-linked one, which is attributed to stronger electronic coupling in the former. These results illustrate the need to carefully tune the electronic coupling between a photosensitizer chromophore and the catalyst to promote photodriven electron transfer yet inhibit adverse electronic effects of the chromophore on electrocatalysis.

Selective oxidative carbonylation of amines to oxamides and ureas catalyzed by palladium complexes

A new process for converting secondary amines into N,N,N?,N?- tetraalkyloxamides under CO pressure, catalyzed by homogeneous palladium complexes in the presence of 1,4-dichloro-2-butene (DCB) as an oxidant, has been developed. The mechanism of the oxidative double-carbonylation process, consisting of the oxidation of Pd(0) to Pd(11) with DCB through a beta-chloride elimination of the eta3-(chloromethyl) allylpalladiuni(11) intermediate, the formation of mono- and bis(carbamoyl)palladium species, and a reductive elimination of the two carbamoyl ligands, is proposed based on studies of the behavior of carbamoylpalladium complexes. When primary amines are employed with DCB as the oxidant, N,N?-dialkyloxamide is catalytically produced, whereas urea is exclusively produced when iodine is used as the oxidant. The reaction of an N-monopropylcarbamoylpalladium complex with propylamine under CO gave N,N?-dipropylurea, whereas a treatment with diethylamine yielded unsymmetrical N,N-diethyl-N?-propylurea, implying the intermediate formation of propyl isocyanate that is converted into the urea upon a reaction with the added amine. A kinetic study on the reaction of chloro-N- propylcarbamoylpalladium with triethylamine suggested a process proceeding through a base-promoted dcprolonalion of the N-monoalkylcarbamoyl ligand to form propyl isocyanate.
