Identifying the biological origin of a disease and the potential targets for intervention are some of the first steps in the discovery of a medicine. The metabotropic glutamate receptor 4 (mGlu4) has lately received much attention due to its potential role in various neuronal diseases such as Parkinson’s disease (PD) and other disorders. Herein we discuss our efforts to develop novel mGlu4 positron emission tomography (PET) radiotracers from mGlu4 positive allosteric modulators (PAMs). We have investigated the biology of this receptor in vitro using cells expressing mGlu4, as well as in vivo, using the 6-hydroxydopamine (6-OHDA)-lesion rat model of Parkinson’s disease. Results of cell studies showed the important role of endogenous glutamate in inducing changes in the affinity of allosteric modulators of mGlus. Results from PET imaging studies showed the interplay between presynaptic mGlu4 and postsynaptic mGlu5 expression in the lesioned side of the brain, while the results of pharmacological MRI studies of the hemodynamic response showed enhanced signal changes in cortical areas in the lesioned side after challenge with mGlu4 PAMs or mGlu5 negative allosteric monitors (NAMs).

The two complexes have electronic spectra dominated by charge transfer, not d-d bands and show no M2+/M 3+ couples. Both complexes show a fluxional 12S3 ligand in solution due to a 1,5-metallotropic shift, an uncommon observation of this particular type of intramolecular ligand exchange. The 195Pt NMR chemical shift of -4201 ppm for [Pt(12S3)2]2+ is consistent with an alternating positioning of the four sulfur lone pairs on the coordinated thioethers. Although 12S3 is poorly pre-organized for facial complexation, its flexibility to position a sulfur in an exodentate fashion enables it to form stable complexes with d8 metal ions such as Pt(II) and Pd(II).

We report the synthesis of one new boronate ester-based covalent organic framework (COF) and two new covalent organic polymers (COPs) made with fluoranthene-containing monomers and hexahydroxytriphenylene. The structure of the monomer heavily influences whether this material forms a highly ordered mesoporous material (COF) or an amorphous, microporous material (COP). The synthesis of the fluoranthene monomers was carried out using a divergent strategy that allows for systematic structural variation and the ability to conduct a careful structure-function study. We found that small structural variations in the monomers dramatically affected the crystallinity, surface area, pore structure, and luminescence properties of the polymers. While each of the monomers contains the same fluoranthene core, the resultant pore sizes range from microporous (10 A) to mesoporous (37 A), with surface areas ranging from ?500 to 1200 m2/g. To help explain how these small structural differences can have such a large effect, we carried out a series of molecular dynamics simulations on the polymers to obtain information with atomic-scale resolution on how the monomer structure affects non-covalent COF layer stacking.
