A pi-acceptor phosphine-electron-deficient olefin ligand was found effective in promoting Pd-catalyzed C(sp)-C(sp) cross-coupling reactions. The new protocol realized the cross-coupling of a broad scope of terminal alkynes and haloalkynes in good to excellent yields with high selectivities. Electron-rich alkynes, which are normally difficult substrates in Glaser couplings, could be employed as either nucleophiles or electrophiles. Alkynes bearing similar substituents, such as n-C5H11CCBr and n-C4H9CCH, which usually suffer from homocoupling side reactions under Cadiot-Chodkiewicz conditions, were successfully cross-coupled in the system. Preliminary kinetic studies revealed that the reaction rate was zero-order in the concentrations of both haloalkynes and terminal alkynes and first order in the loading of Pd(dba)2 and exhibited no obvious dependence on the loading of the copper salt. Control experiments with other phosphines such as PPh3 and DPPF as the ligand were carried out. All the kinetic evidence indicated that the phosphine-olefin ligand facilitated the reductive elimination in the catalytic cycle.

Large-scale synthesis, design, and structure-activity relationships of khafrefungin are reported. Khafrefungin is an antifungal agent that inhibits inositol phosphorylceramide (IPC) synthase, a enzyme involved in fungal sphingolipid biosynthesis. Unlike other inhibitors that inhibit the corresponding enzyme in fungi and mammals to the same extent, khafrefungin does not impair sphingolipid synthesis in mammals. We have developed an efficient method for large-scale synthesis of khafrefungin, and various khafrefungin derivatives were synthesized based on this method. While most of the khafrefungin derivatives lost antifungal activity, a lactone-type derivative had almost the same activity as khafrefungin. We also designed and synthesized derivatives which contain a five-or six-membered ring at the central part of the structure based on NOE experiments of khafrefungin. A macrocyclic khafrefungin derivative was also synthesized, but the antifungal activity was lost. These results suggest that the structure of khafrefungin might be strictly recognized in fungi.

Heterodinuclear complexes, syn-[MPt(mu-pyt)2(bpy) 2]n+ (syn-[MPt], M = Pd2+, Au3+, Hpyt = pyridine-2-thiol, bpy = 2,2?-bipyridine), were synthesized as a selective geometrical isomer by stepwise complexation. X-ray analyses of the hexafluorophosphate salts of these complexes proved their dinuclear structures with short M…Pt distances (2.9084(4) A for syn-[PdPt] and 2.9071(4) A for syn-[AuPt]), similar to the homodinuclear complex (2.9292(2) A for syn-[PtPt]). In the syn-[PdPt] crystal, two dinuclear motifs are arranged closely in a head-to-head manner with a short Pt…Pt distance (3.3757(3) A), forming a dimer-of-dimer structure as in the case of syn-[PtPt], whereas the corresponding crystal of syn-[AuPt] has a discrete arrangement of the dinuclear motifs. By the isomerisation of syn-[PdPt], anti-[PdPt] with equivalent environments of the Pd2+ and Pt 2+ ions was also obtained successfully. Syn-[PdPt](PF 6)2 exhibits vapochromic behaviour based on the absorption/desorption of CH3CN vapour, similar to that observed for syn-[PtPt](PF6)2. The reversible structural transformations induced by the uptake and release of CH3CN molecules were investigated by powder and single-crystal X-ray diffraction studies. These revealed that the vapochromic behaviour was based on the interconversion between two phases, the dimer-of-dimer structure with a short Pt…Pt distance and a pi-pi stacked arrangement with no Pt…Pt intermolecular interaction. The introduction of the heterometal ions enabled control of the colour region: orange ? red for syn-[PdPt] vs. light red ? dark red for syn-[PtPt], reflecting the weaker metal-metal interaction between Pd 2+ and Pt2+ ions in the dinuclear motif. In addition, these complexes were found to exhibit mechanochromic behaviour based on a crystal-to-amorphous transformation upon grinding, and the reconstruction of the crystal structures by vapour sorption.

A series of nickel(II) and palladium(II) complexes [L2M2]2+ have been prepared and structurally characterized, where L is a pyrazolate ligand with bulky 2,6-dimethyl- or 2,6-di(isopropyl)anilinomethyl side arms. Coordinating counter anions such as chloride can bind to axial sites of the dinickel species in a solvent-dependent process, giving rise to five-coordinate high-spin metal ions. In the case of weakly coordinating anions, the metal ions are found in roughly square-planar environments, and the structures are governed by the tendency of the bulky aryl groups to avoid each other, which forces the methyl or isopropyl substituents in the aryl 2- and 6-positions to approach the metal ions from the axial directions. This leads the drastic low-field shifts of the respective 1H NMR signals, e.g. delta = 7.86 ppm for the isopropyl -CH which comes in close proximity to the low-spin nickel(II) center. The relevance of such low-field NMR resonances of protons close to the axial sites of d8 metal ions for possible three-center four-electron M…C-H hydrogen bonds involving the filled dz2 orbital of the metal ion is discussed. In the present case, attractive M…H interactions are assumed to be of no major significance. This was corroborated by the structure of a further [L2Ni2]2+ type complex where the anilinomethyl side arms bear only a single 2-isopropyl group, which was found rotated away from the metal. Additional spectroscopic and electrochemical properties of the various complexes are reported.

Arenediazonium o-benzenedisulfonimides can be used as efficient reagents in Sonogashira coupling reactions. In this work, reactions were carried out in DMSO under very mild conditions (without copper or phosphanes), and gave rise to arylated alkynes in good to excellent yields (25 examples, average yield 83 %). o-Benzenedisulfonimide could be recovered from all the reactions in yields of >80 %, so it could be recycled for the preparation of other diazonium salts. Mechanistic insights revealed the fundamental roles of DMSO and the anion of o-benzenedisulfonimide in the formation of the catalyst, and also the importance of DMSO in the catalytic cycle. The Sonogashira coupling between are diazonium o-benzenedisulfonimides and terminal alkynes is a powerful method for the formation of arylated alkynes. No copper, phosphanes, bases, or co-catalysts are needed. Mechanistic insights highlighted the fundamental roles of DMSO and o-benzenedisulfonimide in the formation of the catalyst, and also the important role of DMSO in the catalytic cycle. Copyright
