Major new advances in synthetic chemistry methods are typically reported using simple, non-standardized reaction substrates, and reaction failures are rarely documented. This makes the evaluation and choice of a synthetic method difficult. We report a standardized complex molecule diagnostic approach using collections of relevant drug-like molecules which we call chemistry informer libraries. With this approach, all chemistry results, successes and failures, can be documented to compare and evolve synthetic methods. To aid in the visualization of chemistry results in drug-like physicochemical space we have used an informatics methodology termed principal component analysis. We have validated this method using palladium- and copper-catalyzed reactions, including Suzuki-Miyaura, cyanation and Buchwald-Hartwig amination.

A concise and convergent total synthesis of the highly cytotoxic marine natural products iejimalide A-D (1-4) is reported, which relies on an effective ring-closing metathesis (RCM) reaction of a cyclization precursor containing no less than 10 double bonds. Because of the exceptional sensitivity of this polyunsaturated intermediate and its immediate precursors toward acid, base, and even gentle warming, the assembly process hinged upon the judicious choice of protecting groups and the careful optimization of all individual transformations. As a consequence, particularly mild protocols for Stille as well as Suzuki reactions of elaborate coupling partners have been developed that hold considerable promise for applications in other complex settings. Moreover, a series of non-natural “iejimalide-like” compounds has been prepared, differing from the natural lead in the polar head groups linked to the macrolide’s N-terminus. With the aid of these compounds it was possible to uncover the hitherto unknown effect of iejimalide and analogues on the actin cytoskeleton. Their capacity to depolymerize this microfilament network rivals that of the latrunculins which constitute the standard in the field. Structural modifications of the peptidic terminus in 2 are thereby well accommodated, without compromising the biological effects. The iejimalides hence constitute an important new class of probe molecules for chemical biology in addition to their role as promising lead structures for the development of novel anticancer agents.

The synthesis of a series of ferrocenylanthracene derivatives is described, utilising the palladium catalysed coupling reaction of 1,1?-bis(chlorozincio)ferrocene with halo-anthracenes. Bis-1,1?-(9-anthracenyl)ferrocene (1) was characterised by single crystal X-ray diffraction and shows an eclipsed ferrocenyl geometry. X-ray crystallographic studies indicate that there are no clear stacking interactions of either an intra-or intermolecular nature between the anthracenyl rings in the structure. A series of 9-and 10-disubstituted ferrocenylanthracene derivatives has also been prepared. In each case the palladium catalyst (Pd(dppf)Cl2) is recovered in a modified form, e.g. as the [(dppf)PdBr(9-anthracenyl)] complex in the synthesis of bis-1,1?-(9-anthracenyl)ferrocene. The single crystal X-ray structure of one such palladium complex [(dppf)PdBr-{9-(10-chloroanthracenyl)}] (15a) has been determined in a case where chloride/bromide exchange had occurred in the palladium complex intermediate. The potential application of compound 1 as synthon for the construction of a molecular sensing device is discussed. Cyclic voltammetry and fluorescence studies have been carried out for selected derivatives.

Synthesis of a novel polycyclic ring scaffold with antimitotic properties via a selective domino Heck-Suzuki reaction

The synthesis of a previously undescribed sp3-rich 6-5-5-6 tetracyclic ring scaffold using a palladium catalysed domino Heck-Suzuki reaction is reported. This reaction is high-yielding, selective for the domino process over the direct Suzuki reaction and tolerant towards a variety of boronic acids.
